Product Description:

NNMTi (5-amino-1MQ) is a potent and selective inhibitor of nicotinamide N-methyltransferase (NNMT), with an IC50 value of 1.2 μM. It binds selectively to substrate-binding site residues of NNMT. NNMTi has shown significant potential in promoting muscle cell differentiation in vitro and enhancing muscle stem cell fusion and regeneration in aged mice.

Mechanism of Action:

    • NNMTi (10-30 μM; 96 hours) induces a concentration-dependent increase in differentiation of C2C12 myoblasts.

In Vivo Studies:

  • Muscle Regeneration in Mice:
    • Subcutaneous injection of NNMTi (5 mg/kg and 10 mg/kg for 2 weeks) significantly improves muscle regeneration post-injury.
    • Results in 60% and 75% higher incidence of proliferating/active muscle stem cells (muSCs) at 5 mg/kg and 10 mg/kg, respectively.
    • The number of fibers with EdU+ myonuclei increases by 40% and 48% at the respective doses.
    • No systemic toxicity was observed even after daily dosing for 1 week, with normal levels of glucose, cholesterol, plasma proteins, and electrolytes.
