Overview
(-)-Huperzine A is a natural alkaloid extracted from the Chinese herbal plant Huperzia serrata. It is a potent, reversible, and highly selective second-generation acetylcholinesterase inhibitor, successfully used in the treatment of Alzheimer's disease.
Currently, (-)-Huperzine A is primarily obtained through plant extraction and chemical synthesis. However, the wild Huperzia serrata resource is nearly depleted due to overharvesting, and the plant is difficult to cultivate on a large scale. Sustainable Huperzia serrata resources are facing severe scarcity. Moreover, chemical extraction and synthesis are complex, environmentally polluting, low-yielding, and costly.
Research Achievements
Our research team has made significant breakthroughs in addressing these challenges. We have cloned 12 enzyme-encoding genes from the endophytic fungus Shiraia sp. Slf14, which is associated with (-)-Huperzine A production. Using these genes, we successfully reconstructed the biosynthetic pathway of (-)-Huperzine A in Saccharomyces cerevisiae. Fermentation experiments confirmed the production of (-)-Huperzine A.
These advancements pave the way for the application of synthetic biology techniques, such as pathway reconstruction and regulatory network engineering, to achieve high-efficiency microbial synthesis of (-)-Huperzine A. This approach is expected to significantly improve fermentation yields and support sustainable, large-scale production.
