Description
Tacrolimus is an important anti rejection drug for organ transplantation, with efficacy 10-100 times that of cyclosporine. It also has good therapeutic effects on immune diseases such as rheumatoid arthritis and lupus erythematosus.
Clinical Applications
Compared with cyclosporine, the use of tacrolimus has completely changed the future of kidney transplantation because it has higher transplant survival rates, higher drug tolerance, lower rejection rates, and fewer side effects. KDIGO recommended tacrolimus as a first-line CNI for renal transplantation in 2009, as an initial and long-term maintenance immunosuppressive therapy. However, the monitoring of tacrolimus concentration is still complex. Insufficient dosage will increase the risk of rejection, while excessive dosage will increase the risk of side effects, mainly nephrotoxicity, neurotoxicity, infection, malignant tumor, diabetes and gastrointestinal discomfort.
